A 2-year postdoctoral fellowship is available at Department of Biotechnology and Biomedicine (DTU Bioengineering), the Technical University of Denmark. The position will be placed in the Enzyme and Protein Chemistry group in the section of Protein Chemistry and Enzyme Technology with about 70 staff members.


The postdoctoral position is part of the project “WPAC. Structure and Design of Whey Protein Alginate Complexes” granted by the Novo Nordisk Foundation.

 

This research is carried out at and coordinated by DTU Bioengineering in collaboration with Department of Biotechnology, Norwegian University of Science and Technology (NTNU), Trondheim; Department of Biology, University of Copenhagen; Center for Molecular Protein Science, Lund University; Department of Food Science, University of Copenhagen; DTU Chemistry; Arla Food Ingredients (AFI) and DuPont. The goal of the project is to advance insights into how the anionic polysaccharide hydrocolloid alginate interacts with important food proteins, such as whey proteins.

 

Responsibilities and tasks

The project work for this position includes production of recombinant b-lactoglobulin and analysis of binding sites interacting with defined alginate oligosaccharides using X-ray crystallography together with DTU Chemistry; enzymatic and chemical cross-linking of whey protein mixtures and identification of cross-linked amino acid residues using mass spectrometry and doing the data analysis with Lund University; analysis of protein-alginate or alginate oligosaccharide complex formation by biophysical techniques (SAXS, DLS, SEC) involving DTU Chemistry, and characterisation of surface properties of protein/alginate complexes as well as their in vitro digestion under simulated gastric and intestinal conditions with Department of Food Science at University of Copenhagen.

 

The project is multidisciplinary and based on collaboration with a group of experts who already collaborate on related research questions. On structural biology and biophysics of protein-alginate oligosaccharide complexes you will be working with P. Harris, G. Peters, J. Ø. Duus and K. Almdal, DTU Chemistry, F. L. Aachmann, Norwegian University of Science and Technology and B. B. Kragelund, SBINLab, Department of Biology, University of Copenhagen. Crosslinking studies and mass spectrometry analysis involves the proteomics platform at DTU Bioengineering and C. Emanuelsson at Department of Biochemistry, Lund University. Analysis of surface properties and in vitro simulated gastrointestinal tract digestion is done with R. Ipsen, Department of Food Science, University of Copenhagen. A PhD student in WPAC uses SPR and ITC for binding analysis with P. Westh, DTU Bioengineering, and is engaged in identification of binding sites on b-lactoglobulin of defined alginate oligosaccharides by NMR titration with SBINLab and NTNU. Several project students work in WPAC.

 

Qualifications
Candidates should have a PhD degree or equivalent.

 

We expect you have a solid background within protein science. Experience in particular with protein structure analysis (X-ray crystallography) as well as some of the below techniques will be an advantage

  • Biophysical methods for measuring protein molecular interactions

  • Recombinant production, purification and characterization of proteins

  • Interactions of oligosaccharides / polysaccharides with proteins and enzymes

  • Protein mass spectrometry

  • Protein structure determination and molecular docking

Salary and terms of employment

The appointment will be based on the collective agreement with the Danish Confederation of Professional Associations. The allowance will be agreed upon with the relevant union.

 

The main workplace will be at the DTU campus in Lyngby, 10 km north of Copenhagen, Denmark. There will be work visits to the partners in Copenhagen, Trondheim and Lund
